I noticed that the eSim on the Galaxy S21 Ultra doesn't pick up the 5GUC. Is the sim card not compatible with T-Mobiles 5GUC? If Samsung does decide to also activate the eSim on the Note 20 will it also not pick up on the 5GUC? I contacted T-Mobile about that issue and I was told that it is something Samsung can fix because the eSim should be able to pick up the 5G UC.

If you have the UC it will pick it up. You might not have that part in your area. Check your coverage map

It has picked UC on the physical Sim I switched back to the physical Sim but it doesn't on the eSim. I think the eSim doesn't have the R13 logo on the eSim that standard physical sims have on T-Mobile.

eSIM is not be supported during beta program on Note 20 devices. eSIM will be supported in an update after official update to Android 12. eSIM should be able to pick up 5G UC if enabled in the area.
