No, the S-pen rely's on a specific type of touchscreen for it to function correctly. It's the same reason why you can't use a Samsung Note series pen on a iPhone or any other phone like the S10 or even an S22 (the S22 works with S-pen). Basically the Z Fold 2 doesn't have the core part to support S-pen.

You can't use an S Pen on either of the Fold 2's screens.

The Fold 3 has the ability to use an S Pen only on the internal (big) screen.
